Comparative evaluation on the therapeutic effect and withdrawal reversing phenomenon of GODEX(R) in hepatotoxicity induced by carbon tetrachloride plus ethanol

Abstract
The protective activity of GODEX¢ç (Hepadif¢ç plus dimethyl diphenyl bicarboxylate [DDB]) against liver injury induced by ethanol plus carbon tetrachloride(CCl4) was investigated in rats. During feeding with ethanol (10%) in drinking water, the animals were orally administered with a high dose (1 ml/kg) of CCl4, followed by repeated low doses (0.5 ml/kg) 3, 7, 10 and 14 days after the 1st administration. The drugs, Hepadif¢ç (250 mg/kg), DDB (30 mg/kg) or their combinational formula GODEX¢ç (250 mg/ kg), were orally treated on days 10, 11, 12, 14 and 17. The reversed increase in serum aspartate transaminase (AST) and alanine transaminase (ALT) levels on 2 days following withdrawal of drug administration after 4-times treatment was very much low in GODEX¢ç-treated rats, although Hepadif¢ç and DDB also exerted effectiveness to some extent. In addition, the rats treated with GODEX¢ç exhibited relatively mild histopathological changes compared with the animals given Hepadif¢ç or DDB. Taken together, it is proposed that the hepato-protective potential of GODEX¢ç, leading to a low withdrawal recurrence of hepatic injuries, is superior to those of Hepadif¢ç or DDB.
